With the ICE Vitrification system, the amount of media that should be loaded into the straw does not matter.  You can load a large column with your cells or a small column.  The cooling rate is not important, therefore a large or small volume will work. 

We suggest you load a 1-2cm column of media with your cells, or approximately 20-40ul.  For oocytes we suggest a slightly smaller column of 10ul of media. 

The media and cells will vitrify in either a large amount or small amount of media.  The cooling rate will be slightly greater with a tiny amount of media but that does not affect survival in this system.  The reason is that the media is very different than other systems.
